Principles:
ATP is the energy currency in all living cells. Its central role makes it necessary for all cells to maintain a constant intracellular concentration, which is similar in all cells. The amount of ATP per cell reflects the intracellular volume and is 1-2 attomoles for bacterial cells and with few exceptions more than 1 femtomole for mammalian cells. Anything interfering with the energy metabolism will lead to a rapid drop in the ATP level and the cell is likely to die within seconds. Consequently the total intracellular ATP amount is a good estimate of the total intracellular volume of the living cells in the sample. If one knows the amount of ATP per cell one can calculate the number of cells per mL. With the Cellular ATP Kit HTS the lysing buffer, an ATPase inhibitor, and the ATP Reagent is combined in one reagent (mix-and-measure). The light emission is extremely stable provided the ATPase activity in the sample is completely inhibited. This makes the kit highly suitable for High Throughput Screening. With some types of cells, e.g. nerve cells, at high cell densities the ATPase inhibitor may not be efficient enough to completely inhibit the ATPase resulting a slowly decaying light emission. This will require using a luminometer with reagent dispensers. Alternatively the Cell Viability Kit SL can be used without dispensers, since the extractant in this kit inactivates ATPase activity completely.
