Principles:

The genetically fused biotinylated recombinant thermostable luciferase can be linked via streptavidin to any biotinylated primary or secondary antibody. After washing and addition of luciferin and ATP, luminescence output can be immediately recorded in a luminometer. This is a highly sensitive assay with a detection limit of 10-20 mol luciferase.


An even higher sensitivity can be obtained by using biotinylated acetate kinase (not commercially available). ADP + acetyl phosphate -----> acetate + ATP. Bioluminescent detection of the ATP by the firefly luciferase enzyme can be used to detect the marker enzyme at a detection limit of 10-21 mol.

Kinase kit for HTS applications in Drug Development

Our new kit uses direct measurement of ATP degradation rate to distinguish hits from non-hits, in drug development. The methodology is flexible and can easily be adapted to various HTS strategies.
